Anti-CD40L Monoclonal Antibody Overcomes CD4-Mediated Graft Rejection

Treatment with anti-CD40L monoclonal antibody (MAb) prevents CD4 T-cell mediated resistance to allogeneic bone marrow engraftment.

"We previously reported that costimulatory blockade can eliminate the need for thymic irradiation (TI) or repeated administration of T-cell-depleting MAbs (TCD MAbs) in an allogeneic bone marrow transplant (BMT) regimen for inducing specific tolerance," said H. Ito and colleagues from the Massachusetts General Hospital. "Here we analyze the role of host CD4 T cells and CD8 T cells in resisting the induction of mixed chimerism in mice receiving anti-CD40L MAb."
