Suicide Gene Therapy

"Retrovirally Transmitted Gene Therapy for Plasma Cell Tumors using Herpes Simplex Virus Thymidine Kinase Gene."

According to an abstract submitted by the authors to the Keystone Symposia on Molecular and Cellular Biology entitled Gene Therapy for Hematopoietic Stem Cells in Genetic Disease and Cancer, held February 4-10, 1996, in Taos, New Mexico, "Human myeloma cells of the ARM-77 cell line were transduced with the herpes simplex thymidine kinase (HSVtk) neomycin phosphotransferase fusion gene using the PLNT 3.3 vector. The cells were injected s.c. on four locations on the back of SCID mice in the following proportions between transduced and nontransduced cells: 100%/0%, 50%/50%, 25%175%, 0%/100%. One week later ganciclovir (GCV) was given i.p. in different concentrations for 2 or 3 weeks. ...
