Tumor Amplified Protein Expression Therapy with Salmonella Vector

Suicide gene therapy for cancer may travel via a new vector. Researchers from Vion Pharmaceuticals and Yale University School of Medicine, Connecticut, have recently genetically engineered a non-pathogenic strain of Salmonella typhimurium to be hyperinvasive to tumors in vitro and in vivo (see other stories in this issue). When injected into tumor bearing mice, these attenuated bacteria rapidly home in on tumors, while ignoring normal tissues. Tumor:liver ratios as high as 10,000:1 have been achieved, depending on the tumor type, and the bacteria themselves have been shown to have an anticancer effect.
