Cell-Specific Transgene Expression Prevents HSVtk/GCV Toxicity in the Liver

Tumor cell-specific transgene expression prevented liver toxicity that is normally associated with adenovirus-mediated herpes simplex virus thymidine kinase (HSVtk)/ganciclovir (GCV) suicide gene therapy.

Suicide gene therapy has undergone intense scrutiny as a potential anticancer therapeutic. The HSVtk/GCV system has received the most study because the efficacy of this approach is enhanced by the bystander effect. However, the treatment of malignancies with an adenovirus (Ad) vector can lead to the transduction of cells that are not cancerous.
