Membrane Permeable Purines Induce Efficient Bystander Killing

A recent study showed that tumor cells transfected with the Escherichia coli (E. coli) purine nucleoside phosphorylase (PNP) gene produced a 100 percent tumor killing effect when as few as 2% of the tumor cells were successfully transfected.

Selective expression of therapeutic genes within tumor cells is a required property for the successful use of gene transfer to sensitize tumors to specific chemotherapies. One flexible method of tumor targeting is the application of tissue- or tumor-specific promoters to encourage toxin gene expression within a specific target tissue.
