Suicide gene therapy using the adenoviral-mediated herpes simplex thymidine kinase (Ad.tk) gene transfection method followed by ganciclovir (GCV) treatment shows promise as a primary treatment for cancers and as well as a secondary therapy for destruction of residual cells. Its use has also been documented in the purging of autologous donor materials of cancer cells before retransplantation.
Unfortunately, host immune response against the adenoviral vector most often used in this therapy reduces the efficacy and decreases the window of effectiveness....
