"A Rational Strategy for the Design Of Anti-HBV Nucleosides."

According to an abstract presented by the authors to the 1st International Conference on Therapies for Viral Hepatitis, held December 11-15, 1995, in Kauai, Hawaii, "Dideoxyribonucleoside (ddN) analogues are widely used as chemotherapeutic agents towards various viral infections. Like the constitutive deoxynucleosides (dN), such ddN must be metabolized to their 5'-triphosphate analogue (ddNTP) before exerting their biological effect. More precisely, the ddNTP/dNTP ratio is one of the most important determinants of any antiviral nucleoside activity as the ddNTP competes with endogenous dNTP for binding to the viral polymerase or acts as an alternate substrate.
