Liver Toxicity Associated with Adv-Mediated HSV-tk/GCV Gene Therapy

Liver-associated toxicity was realized in a murine model of herpes simplex virus thymidine kinase/ganciclovir (HSV-tk/GCV) suicide gene therapy mediated by adenoviral vectors.

Animal models with artificially induced liver tumors of colorectal origin treated locally with either retrovirus or adenovirus vector-mediated HSV-tk/GCV suicide gene therapy have been studied previously. Although extensive testing of retroviral vectors has not revealed any major signs of toxicity (D.W. Moorman, J. Surg. Oncol., 1994;57:152-6), no toxicity studies using adenoviral mediated HSV-tk/GCV gene therapy have been reported to date.
