E3 Overexpression Prevents Immune Responses Against Vector

Overexpression of the immunomodulatory product of the early adenovirus genome at region 3 prevents the host antiviral immune response that limits the effectiveness of adenoviral vectors in gene therapy protocols.

Although highly efficient at transferring foreign genes to the liver, as well as to quiescent cells, recombinant adenoviruses (Ads) have a limited duration of gene expression due to both humoral and cellular immune responses by the host against the vector. This antiviral immune response also prevents re-administration of the vector if additional treatment is necessary.
