According to an abstract submitted by the authors to the 6th Symposium on Gene Therapy: Towards Gene Therapeutics, held May 4-6, 1998, at the Max Delbruck Center for Molecular Medicine, Berlin-Buch, Germany, "The delivery of a therapeutic gene as an episome offers a potential solution to eliminate position effects and alleviate size constraints. To shuttle therapeutic genes as self-replicating DNA in human cells, non-infectious and infectious episomal vectors based on the latent human herpes Epstein-Barr virus (EBV) have been developed (Nature Genetics 8:33; Nature Medicine 1:1303; Human Gene Therapy 6:1005; Gene Therapy 3:1081), including systems for shuttling large inserts from YAC...
