To maximize the efficiency of DNA delivery during gene therapy, M.T.G. DaCruz and colleagues studied the factors involved in binding, association, and fusion of lipoplexes and cells. Effects of stoichiometry of cationic lipids and DNA, the presence of serum in the cell culture medium, and the incorporation of the ligand transferrin into the lipoplexes were examined. As a control, conditions were mimicked in experiments including endocytotic inhibitors.
