Nested PCR Method Detects Rifampin Resistance In Sputum Smears

Saltmarsh, staff medical writer - Researchers in South Korea have overcome hurdles that prevented the detection of rifampin resistance in sputum smears by using a new, nested polymerase chain reaction (PCR) method that can specifically amplify the rpoB DNA of Mycobacterium tuberculosis, according to a report in the Journal of Clinical Microbiology.

"Either PCR-mediated single strand conformation polymorphism (SSCP) analysis or DNA sequencing of rpoB DNA (157 bp) can be used as a rapid screening method for the detection of mutations related to the rifampin resistance of M. tuberculosis," reported B.J. Kim and colleagues at Seoul National...
